Label-free Quantification of Intracellular Lipids

thum_resources_03.png

Lipid droplets have distinctly high refractive index values, making them naturally visible in holotomographic images without any staining. HT enables label-free 3D quantification of lipid content in live cells, supporting time-lapse studies of metabolic dynamics. Applications span liver disease research, metabolic studies, and biofuel-relevant microalgae biology.

- Label-free 3D imaging and quantification of lipid droplets in live human hepatocytes (Huh-7), validated against Nile Red staining
- Measurement of lipid droplet volume, mass, and distribution following oleic acid treatment
- Lipid content quantification in single microalgae (Nannochloropsis oculata) under nitrogen deficiency
- Correlative HT and fluorescence imaging using HT-2H for co-localization studies
